will I have to change the gearing in my rustler vxl 2wd to use these with a 3s lipo?

It's always difficult to advise on gearing as it can change based on your driving style and the surface you are running on. However, yes - it's likely you'd want to drop a tooth or two on your pinion when running these on 3S.

The ProLine Badlands MX28 Tyres are listed as having front and rear versions for the Traxxas Rustler 4x4. My question is what is the difference since the images and specs appear to be identical and the stock wheels on the Rustler 4x4 are the same front and back?

The offsets are different on the wheels , with the 4x4 you can use the same all round.
